Germline mutations in MSH6, a DNA mismatch repair gene, increased the risk of colorectal cancer by 8-fold, the risk of endometrial cancer by 26-fold, and the risk of other cancers associated with Lynch Syndrome by 6-fold compared to the general population.

A comparison of sporadic colorectal cancer with microsatellite-instability (MSI) high pathology indicated that distal MSI-high tumors were associated with differentiated morphology, distant metastasis, and significantly worse 3-year disease-free survival and overall survival compared to proximal MSI-high tumors.

In a retrospective analysis of the PETACC-3 adjuvant chemotherapy trial in patients with stage II and III colon cancer, KRAS mutation status was not of major prognostic value for relapse-free or overall survival, but BRAF mutation status was prognostic for overall survival in the patients with microsatellite instability low and stable tumors.

In the randomized phase II CELIM trial, neoadjuvant chemotherapy plus cetuximab produced high response rates compared to historical controls and resulted in a significant increase in tumor resectability in patients with metastatic colorectal cancer and baseline unresectable liver metastases.
