Purpose

In spite of the high response rates yielded by imatinib, the current standard of care for the first-line treatment of patients with chronic myeloid leukemia (CML), there is a high rate of relapse. Furthermore, some patients with advanced stages of the disease develop resistance to imatinib. Studies have identified mutations in the BCR-ABL tyrosine kinase (TK) domain that are responsible for resistance to imatinib, leading to the development of newer agents that might overcome this resistance. Several novel TK inhibitors (TKIs) have recently gained US Food and Drug Administration approval for the treatment of imatinib-resistant/refractory CML. The purpose of this activity is to update physicians on the current issues in the treatment of CML and apprise them of the current recommendations for the optimal management of CML.

Target Audience

This activity is intended for medical oncologists and hematologists involved in the care of patients with chronic myeloid leukemia. No specific skills or knowledge other than a basic training in oncology are required for successful participation in this activity.

Learning Objectives

Upon completion of this educational activity, you should be able to:

  • Summarize the various testing options for monitoring response to therapy with small-molecule inhibitors of the Bcr-Abl tyrosine kinase in patients with chronic myeloid leukemia in chronic phase
  • Discuss the efficacy of second-generation Bcr-Abl tyrosine kinase inhibitors in patients with imatinib-resistant or -intolerant chronic myeloid leukemia in chronic phase
  • Analyze the management of adverse events, especially myelosuppression, in patients with chronic myeloid leukemia receiving therapy with second-generation Bcr-Abl tyrosine kinase inhibitors
