Treatment options for castration-resistant prostate cancer (CRPC) are limited, with a modest 3-month improvement in median overall survival with docetaxel-based chemotherapy, the current standard of care. At disease progression, there is no accepted salvage therapy. Patients frequently suffer significant and debilitating comorbidities resulting from the propensity of prostate cancer cells to metastasize to bone. Recently, a variety of agents, including epothilones, vaccines, endothelin A receptor antagonists, and androgen synthesis or signaling inhibitors, have shown activity in both first and subsequent lines of therapy for CRPC. A novel RANKL (receptor activator of nuclear factor-κB ligand) inhibitor has demonstrated efficacy in preventing bone complications in patients with prostate cancer. Clinical development of these agents is continuing in ongoing phase III trials.
The purpose of this activity is to update physicians on emerging therapies and ongoing clinical trials for the treatment of patients with advanced CRPC.
