Summary and Purpose

An increased understanding of the molecular defects in lung cancer has led to the development of novel agents to target specific biologic pathways. Targeted agents have been approved by the US Food and Drug administration in combination with platinum-based chemotherapy in the first-line treatment of patients with advanced non-small-cell lung cancer (NSCLC) and as single agents in pretreated metastatic NSCLC. While avoiding the myelotoxicity typically associated with cytotoxic chemotherapy, these targeted agents are associated with their own stereotypic class effects that are beginning to be characterized, which will necessitate the development of new supportive care measures. Additionally, differences in DNA repair and nucleotide metabolism might affect a tumor cell's sensitivity to different cytotoxic agents, and the choice of chemotherapy regimen might be optimized according to these characteristics in future regimens.

The purpose of this activity is to apprise physicians on advances in the treatment of patients with NSCLC.

Target Audience

This publication is intended for medical oncologists involved in the care of patients with non-small-cell lung cancer. No specific skills or knowledge other than a basic training in oncology are required for successful participation in this activity.

Learning Objectives

Upon completion of this educational activity, you should be able to:

  • Compare the significance of different clinical and molecular markers in the pathogenesis of NSCLC.
  • Summarize the findings of clinical trials and retrospective analyses to correlate molecular patient and tumor characteristics to clinical outcomes in NSCLC.
  • Identify the different strategies that can be utilized to optimize responsiveness to inhibition of epidermal growth factor receptor in NSCLC
  • Analyze the safety and efficacy of antiangiogenic agents alone and in combination with chemotherapy in NSCLC
  • Describe the adverse events associated with targeted therapy in NSCLC
  • Describe the safety and efficacy of novel therapeutic agents in NSCLC
